Nuclei isolation and fluorescence activated nuclei/cell sorting (FANS/FACS). Preparation of single-cell methylome (snmC-seq2) and multi-omic (sn-m3C-seq, snmCT-seq) libraries using a robotic liquid handler system (Beckman Biomek i7). Prepare buffers and solutions. Manage the inventory of reagents and tissue samples used for single-cell experiments. Perform quality control analyses of high-throughput sequencing libraries. Accept responsibility for performance of all laboratory procedures under the direction of supervisors. Keep an up-to-date standard operating procedures (SOPs). Maintain freezers and refrigerators by periodic thawing and cleaning and organizing their contents Manage biohazard and recycle wastes. Ensure adequate supply of liquid nitrogen for cell storage and CO2 for tissues culture. Monitor equipment function and report and arrange for repair of malfunctioning equipment. Clean the tissue culture hood, incubator and surfaces as well as pipettes and pipet aids as needed.
